ARSACS research has traditionally focused on neurons, particularly Purkinje cells in the cerebellum. However, growing evidence suggests that microglia, the immune cells of the brain, may also contribute to the disease. Recent findings indicate that abnormal microglial activity may appear very early in ARSACS, even before significant neuronal loss occurs. This project will investigate whether microglia are simply reacting to neuronal damage or actively contributing to disease progression. Researchers will combine two complementary models: microglial cells and neurons generated from stem cells derived from ARSACS patients, and a genetically modified zebrafish model that allows microglia and neurons to be observed in the living brain. The team will study inflammation, metabolism and cell interactions, and will also test therapeutic approaches designed to reduce harmful inflammation and protect neurons. The findings could reveal new mechanisms and therapeutic targets for ARSACS.
